Defective Phage Formation by Lysogens of Integration

چکیده

Integration deficient (L) mutants of phage P22 can be complemented with L+ phage to yield L mutant lysogens. Once established the L prophage is stable in the absence of L+ function. Ultraviolet or thermal induction of L lysogens leads to product, ion of defective phage particles and low yields of infectious phage. The defective particles contain a normal amount of DNA but the DNA is mainly bacterial in origin and appears to derive from the pro C region adjacent to one end of the prophage. The defective lysates t,ransduce pro C at high frequency relative to other bacterial markers. The representation of phage genes is strongly polar with markers distal to pro C being rarely present. It appears that L+function is required not only for efficient integration of the prophage, but for normal recovery of the prophage genome following induction.

Some improved methods in P22 transduction.

Recent refinements simplify methods for P22 transduction in Salmonella and allow improved recovery of phage-free transductional clones. The methods include use of: integration- and lysis-defective phage mutants, heat-killed bacteria to eliminate free phage, direct plating of phage and bacteria, replica-plating for detection of phage content of individual clones, improved broth for phage growth,...

Relationship between prophage induction and transformation in Haemophilus influenzae.

The interaction between transformation and prophages of HP1c1, S2, and a defective phage of Haemophilus influenzae has been investigated by measurement of (i) the effect of prophage on transformation frequency and (ii) the effect of transformation on phage induction. The presence of any of the prophages does not appreciably alter transformation frequencies in various Rec(+) and Rec(-) strains. ...

Mapping of integration and excision crossovers in superinfection double lysogens for phage lambda in Escherichia coli.

I N a previous paper it was shown that one prophage apparently acts as a homology region for a superinfecting phage of the same kind (CALEF, MARCHELLI, GUERRINI 1965). A more rigorous test for the same possibility can be obtained by correlating the frequency of integration of superinfecting phage at any given region with the map length of that region. The work presented here consists primarily ...

Mycobacteriophage L 5 Integrase - Mediated Site - Specific Integration In Vitro MONG

Mycobacteriophage L5, a temperate phage of the mycobacteria, forms stable lysogens in Mycobacterium smegmatis via site-specific integration of the phage genome. Recombination occurs within specific phage and bacterial attachment sites and is catalyzed by the phage-encoded integrase protein in vivo. We describe here the overexpression and purification ofLS integrase and its ability to mediate in...

Effect of glycerol on Haemophilus influenzae transfection.

Competent Haemophilus influenzae bacteria were exposed to purified phage HP1 DNA and then plated for transfectants (PFU). When 32% (final concentration) glycerol was added before plating, between 10- and 100-fold more transfectants were observed. Glycerol had no significant effect on transfection with DNA from single or tandem double lysogens. It also had little effect on transformation with ch...
